Rune Mysteries is a fundamental OSRS Free-to-Play quest that must be completed to gain access to the Runecrafting skill. Started by speaking to Duke Horacio in Lumbridge Castle, the quest involves acting as a messenger. You deliver a Strange Talisman from the Duke to Head Wizard Sedridor in the Wizards' Tower basement. Sedridor then gives you research notes to take to Aubury, the rune shop owner in Varrock. Aubury gives you his own research notes to return to Sedridor, completing the quest and unlocking the ability to train Runecrafting.
